A Pomegranate Compound Shields Skin Cells From UV Damage by Activating the Body's Own Antioxidant Defense System

UVA radiation, the longer-wavelength component of sunlight that penetrates deeply into the skin, generates large amounts of reactive oxygen species (harmful free radicals) inside skin cells, leading to oxidative stress, mitochondrial damage, cell death, and over time the breakdown of collagen and accelerated skin aging. This 2024 laboratory study by researchers at Università Cattolica del Sacro Cuore in Rome, published in Current Research in Pharmacology and Drug Discovery, investigated whether punicalagin, a powerful polyphenolic compound found primarily in pomegranates, could protect human dermal fibroblasts (the skin's structural cells) from this UV-induced oxidative damage. When skin fibroblasts were treated with punicalagin and then exposed to UVA radiation, the compound produced significant protective effects across multiple biological measures: it increased cell survival, dramatically reduced the levels of damaging reactive oxygen species generated by UV exposure, and critically, activated the Nrf2 pathway, a master regulatory switch in human cells that controls the body's own internal antioxidant defense system. Activation of Nrf2 in turn triggered increases in reduced glutathione and NADH, two of the body's most important antioxidant molecules, and protected the mitochondria (the cell's energy-producing structures) from UV-induced damage, preserving their respiratory capacity. The compound also demonstrated anti-apoptotic effects, meaning it helped prevent skin cells from undergoing programmed cell death in response to UV damage. Together these findings suggest that punicalagin does not simply act as an external antioxidant scavenger but works more powerfully by switching on the cell's own protective machinery, making it a promising candidate for use in both dietary supplementation and topical skin care formulations aimed at UV protection and anti-aging.

Abstract

Polyphenols are a class of natural compounds that act as antioxidants, neutralising harmful free radicals that would damage cells and increase the risk of diseases such as cancer, diabetes and heart disease. They also reduce inflammation, which is thought to be at the root of many chronic diseases. We are investigating the photoprotective effects of punicalagin, a type of polyphenolic compound mainly found in pomegranates, against UVA-induced damage in human skin fibroblasts. Punicalagin increases cell viability and reduces the high levels of ROS generated by photooxidative stress through its ability to modulate the Nrf2 transcriptional pathway. Interestingly, activation of the Nrf2 pathway results in an increase in reduced glutathione, NADH, and subsequently protects mitochondrial respiratory capacity. Integrating molecular and imaging approaches, our results demonstrate a potential cytoprotective effect of punicalagin against UVA-induced skin damage through an anti-apoptotic mechanism.
